Description

Description: This assay employs a two-site sandwich ELISA to quantitate CDH2 in samples. An antibody specific for CDH2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CDH2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CDH2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CDH2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Cadherin 2, type 1, N-cadherin (neuronal), is a classical cadherin from the cadherin superfamily. The encoded protein is a calcium dependent cell-cell adhesion glycoprotein comprising five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. The protein functions during gastrulation and is required for establishment of left-right asymmetry. At certain central nervous system synapses, presynaptic to postsynaptic adhesion is mediated at least in part by this gene product. Reid and Hemperly (1990) obtained a full-length NCAD clone from a Kelly neuroblastoma library. The NCAD gene encodes a 907-amino acid protein that includes a 159-amino acid signal sequence. Human and mouse nucleotide sequences are 96% identical
